This thesis develops analytical models of congested networks based on finite capacity queueing theory. These models are then used to evaluate and identify traffic management strategies that mitigate congestion in three different application fields: health care, biology and urban transportation. This thesis also proposes a simulation-based optimization (SO) framework that combines information from a detailed, yet computationally inefficient, stochastic traffic simulator with information from an efficient, yet less realistic, analytical traffic model. This combination leads to an SO framework that achieves an excellent trade-off between detail and tractability. The framework is used to solve an urban traffic signal control problem for the Swiss city of Lausanne.
